Bladder cancer is the fifth leading cancer in Germany. About 30% of the cases are muscle invasive (MIBC) at first diagnosis, requiring radical cystectomy. The novel photosensitizer Tetrahydroporphyrin-Tetratosylate (THPTS) is excitable at near-infrared wavelength of 760nm) and may thus be suitable for Photodynamic Therapy (PDT) of MIBC up to 15 mm thickness. We tested the tumoricidal effect of THPTS-PDT in an orthotopic animal model, and explored cellular mechanisms of action in cultured cells. Single PDT of invasive bladder cancer led to a significant reduction of tumor size and pT-grade at 14 days after treatment. Apoptosis was the predominant modality of cell death. In addition, we found an immunostimulatory effect of THPTS-PDT in the rat model.
